Avonturier Nathan Drake keert terug in Uncharted 2: Among Thieves. Het verhaal speelt zich enkele jaren na Uncharted: Drake's Fortune af. Wederom dient er een historisch mysterie opgelost te worden en deze keer duikt Nathan in het mysterie rondom Marco Polo. Na twintig jaar bij de keizer Kublai Khan doorgebracht te hebben, vertrekt Marco Polo in 1292 huiswaarts met 14 schepen en ruim 600 passagiers aan boord. Als hij echter anderhalf jaar later aankomt in Portugal is er nog maar één schip met 18 passagiers over. Marco Polo heeft nooit een woord gesproken over de verdwenen schepen. In Uncharted 2: Among Thieves kruipt de speler in de huid van Nathan om weer en wind, maar ook vele vijanden te trotseren in zijn zoektocht naar een historisch artefact. Al klimmend, rennend en springend baant hij zich een weg door verschillende exotische locaties. Nathan is uitgerust met een ruim arsenaal aan wapens en vechttechnieken om de vele vijanden die op zijn pad wachten, te verslaan. Naast de uitgebreide singleplayer-modus is het verhaal van Uncharted 2: Among Thieves ook met zijn tweeën door te spelen, terwijl spelers het in de verschillende multiplayermodi met zijn tienen tegen elkaar op kunnen nemen. Zo bevat de game een Team Deatchmatch-modus, waarin twee teams van vijf spelers elkaar het leven zuur maken, terwijl het in de Capture the Treasure-modus draait om het veroveren en veilig stellen van een in het level verstopte schat.
